Abstract

Colon targeted drug delivery in cancer chemotherapy is advantageous as it increases drug concentration at the site of action (colon), reduces extensive first pass metabolism, which ultimately reduces adverse effects caused by conventional chemotherapy. Fast dissolving tablets-in-capsule system has added advantage as it is easy to swallow, denser compared to pellets, slippery, hence takes less time to reach colon over variable GIT transit time. Hence, advanced drug delivery system is needed which benefits in the treatment involving cancer chemotherapy. Regorafenib is widely considered as drug of choice in treatment of colon cancer, even though there is formation of toxic metabolites through first pass metabolism in liver causing hepatotoxicity which is sometimes reported fatal. The dosage form was designed to overcome liver toxicity due to first pass metabolism and to effectively use in the treatment of colon cancer, by reducing adverse effects and considerably eliminating fatality rate in cancer chemotherapy. This project creates a path for the future for upgrading technology in treatment of cancers.
