Abstract

BACKGROUND: Cell migration is an essential biological process that regulates both development and disease. One of the factors known to regulate cell migration is the actin-binding protein, Fascin. It is an actin-bundling protein that cross-links individual actin filaments into straight and stiff bundles and it is typically thought to promote cell migration through bundling actin to form migratory structures such as filopodia and invadapodia. Fascin overexpression in cancer is strongly associated with poor prognosis and metastatic progression across different cancer types. According to numerous literature evidence, it is believed that Fascin plays a causative role in promoting metastatic progression, but thorough literature search provided very few reports with this marker in oral squamous cell carcinoma. With this background, we conducted this study to evaluate the role of Fascin in oral squamous cell carcinoma. AIM OF THE STUDY: The aim of this study was to determine the Expression of Fascin in different grades of oral squamous cell carcinoma using Immunohistochemistry. OBJECTIVES: 1. To analyse the intensity of staining of Fascin in normal oral mucosa, well differentiated oral squamous cell carcinoma, moderately differentiated oral squamous cell carcinoma and poorly differentiated oral squamous cell carcinoma. 2. To analyse the area of staining of Fascin in normal oral mucosa, well differentiated oral squamous cell carcinoma, moderately differentiated oral squamous cell carcinoma and poorly differentiated oral squamous cell carcinoma. 3. To compare the intensity of staining of Fascin expression in normal oral mucosa, well differentiated oral squamous cell carcinoma, moderately differentiated oral squamous cell carcinoma and poorly differentiated oral squamous cell carcinoma. 4. To compare the area of staining of Fascin expression in normal oral mucosa, well differentiated oral squamous cell carcinoma, moderately differentiated oral squamous cell carcinoma and poorly differentiated oral squamous cell carcinoma. MATERIALS AND METHOD: A total of 40 paraffin embedded tissue blocks were retrieved from the archives of the department of Oral Pathology and Microbiology. The study sample consisted of ten each of previously diagnosed well differentiated oral squamous cell carcinoma(WDSCC), moderately differentiated oral squamous cell carcinoma(MDSCC), poorly differentiated oral squamous cell carcinoma(PDSCC)and Normal oral mucosa(NOM). The tissue blocks were sliced into 3 μm thick sections and tissue sections were mounted on positively charged slides. Immunohistochemical staining of Fascin was performed and the immunoreactivity of the marker was analyzed. Evaluation of Fascin tumor marker was done using selected fields rich in staining. The data obtained were statistically subjected to SPSS software version 20.0. RESULTS: On Comparing intensity of staining and area of staining of Fascin among Group I (NOM) with all other groups, Group-II (WDSCC), Group-III (MDSCC) and Group-IV (PDSCC), statistically significant values were obtained. On comparing Group II (WDSCC) with Group III (MDSCC) and Group IV (PDSCC), p values were found to be statistical significant, whereas while comparing Group III (MDSCC) and Group IV (PDSCC), a p value of 0.30 was obtained which showed no statistical significance. CONCLUSION: From the present study, we conclude that Fascin shows significant expression in varying grades of oral squamous cell carcinoma. This could imply that Fascin can be used as a prognostic marker for the prediction of prognosis of oral squamous cell carcinoma. The analysis of Fascin expression may also help in early detection of tumors there by reducing the mortality and morbidity of oral cancer.
