Abstract

The present studies were aimed at developing Simvastatin solid dispersion using combination of polymers like PEG6000:PEG4000 prepared by using solvent evaporation technique. The simvastatin solid dispersion were evaluated for physicochemical characteristics like drug content and in-vitro drug release studies. From FTIR spectroscopy studies, it was concluded that there was no defined chemical interactions between Simvastatin and PEG4000, PEG6000. It can provide a promising way to enhance its solubility and dissolution rate. The compatibility of the drug and excipient is confirmed by carried out the DSC and XRD analysis which clearly shows compatibility of the polymer and drug. The optimized formulation F1 was compared with marketed product, the results were found that formulation F1 shows in vitro drug release 86.95% in 30min and marketed product shows 80.08% in 30 min. Experience with solid dispersions indicates that this is a very fruitful approach to improve the release rate and oral bioavailability of poorly soluble drugs. Based on mathematical models, it was concluded that formulation F1, the regression value was found to be 0.98 fitted into zero order release kinetics.
