Abstract
Thin layer chromatography of thespesia populnea leaf extract showed the presence of quercetin the Rf value is compared to standard quercetin. HPTLC result of thespesia populnea leaf extract explained the quantitative estimation of quercetin content . The amount of Quercetin in Hydro alcoholic extract of Thespesia populnea was found to be 0.1972%w/w. After confirmation of presence of quercetin from thespesia populnea leaf extract then isolate the quercetin by using column chromatography. Repeated the column isolation got a high yield of quercetin. The isolated quercetin was characterized by UV-Visible , FT-IR, NMR, LC-MS spectroscopy .The spectroscopic results explained the isolated quercetin structur and molecular weight. The swiss target prediction was done by isolated quercetin structure to find out the receptor binding efficiency in biological membrane. Small quantity of Isolated quercetin was used as capping and stabilizing agent to synthesis the copper nanoparticles by green chemistry method . Greenly synthesized copper nanoparticles was characterized by UV-Visible ,FTIR ,AAS Spectroscopy and SEM,EDAX,XRD,ZETA POTENTIAL , PARTICLE SIZE Analysis.potential than isolated quercetin is confirmed by the IC50 values in different pharmacological evaluation so further development needed to make a copper nanoparticles in anticancer therapy . In normal charged copper ion itself having a mild anti-cancer agent and more toxic if the charged copper is converted into copper neutral ion or copper nanoparticles had less toxic and more active. The reduction of charged copper ion to neutral copper ion by chemical method, more chemical agent needed and sometime may be form some side reaction and produce a toxic product. If natural product used to the reduction process of copper ion form a good neutral copper atom or copper nanoparticles . In this study isolated flavonoid (natural product) used as an reducing and stabilizing the copper sulfate into copper nanoparticles .it was environmental friendly and nontoxic method for synthesis of copper nanoparticles. In future development of copper nanoparticles green chemistry based synthesis is suitable for non toxic formulation and cancer target delivery.
