Abstract

The present study is an attempt to investigate the effect of Aragwadhadi Kashayam on Alloxan induced diabetics in wistar albino rats. Qualitative phytochemical screening and ethno botanical survey on Aragwadhadi kashyam shows the presence of certain phyto constitutions such as proteins, amino acids, tannins, flavonoids, carohydrates, glycosides and terpenoids. The phytoconstituents such as glycosides, tannins, alkaloids, triterpenoids and flavonoids may be linked to the anti diabetic activity. Alloxan treatment will produce significant increase in serum glucose level with respective normal control group. The administration of drug 200mg/kg and glibenclamide 5mg/kg significantly reversed the increase in serum glucose level. Diabetic mellitus (DM) is an endocrine disorder in which the glucose metabolism is impaired because of total loss of insulin after destruction of pancreatic beta cells or because of inadequate release of insulin from the pancreatic cells of beta cells. The fundamendal mechanism underlying hyperglycemia involved over population and decreases utilization of glucose by the tissue. Dose dependent effect of the glibenclamide showed rapid normalization of blood glucose due to its insulin releasing effect. In our present study there was a significant weight gain formulation treated the diabetic rats compared with the normal control rats and this observation shows anabolic effect of the formulation on body weight on the diabetic rats. Hyperglycemia and insulin resistance both seem to have important in the pathogenesis of macro vascular compplications. Diabetes mellitus causes a disturbance in the uptake of glucose as well as glucose metabolism. The hyperglycemia in the diabetes might inhibit tissue repair in the macro vascular beds. In the present study of formulation treated group shows hypoglycemic activity and it confirms the presence of the anti- diabetic activity. Sulfonylurea such as glibenclamide is often used as a standard drug in the Alloxan induced diabetic to compare to the efficacy of antihyperglycaemic compound. In study there was a significant elevation in blood glucose level in the diabetic control group as compared with normal animal. The formlation treated group exhibited significant reduction of fasting plasma glucose level as compared to the diabetic control group. Over production of glucose by means excessive hepatic glycogenolysis and gluconeogenesis is one of the fundamental basis of hyperglycemia in diabetes mellitus. Diabetes is a common chronic ailment for which the patient has to take insulin to maintain the blood sugar level. It is interesting to see how the formulation tackles this problem. It corrects the function of pancreas, stimulating it to produce insulin in the natural way, which in turns way to maintain the blood sugar level. Extract revitalizes and rejuvenates the organs, the dysfunction of which is causing the disease. This bring back normal functioning of the organs. It is also maintaining the healthy state body. Since no artificial chemical are involved, it doesn’t cause any side effects. Conclusion The presented study is an attempt to investigate the effect of Aragwadhadi kashayam on Alloxan induced diabetes in wistar albino rats. The Preliminary Phytochemical study was done which showed the presence of tannins, carbohydrate, flavonoids and reducing sugar which is responsible for the anti- diabetic activity. The animals were given Alloxan at a dose of 120mg/kg intraperitonially and the diabetic animals were treated with drug Aragwadhadi kashayam at a dose of 200mg/kg and 400mg/kg for 21 days. The serum glucose, body weight, levels of Liver function tests, Haemoglobin content, Post Prandial glucose levels, AST, ALP, ALT, Urea, Creatinine, Total Bilirubin were significantly improved when compared with positive control group. All these observation imply that the Aragwadhadi kashayam could be regarded as an favorable Anti-diabetic agent. The present investigation suggests that the Aragwadhadi kashayam has potential as an anti diabetic agent against the Alloxan induced diabetes. The mechanism of action of the drug is not completely established which needs further studies.
