The test drug Kukkilathi Urundai was selected from the siddha literature Anuboga Vaithiya Navaneetham to proof the scientific evidence. The dissertation started with the introduction that explain about the basic siddha concept, shortage of scientific validation on siddha drugs and prevalence of non-healing ulcers and role of test drug in treating non-healing ulcers. All the ingredients for this trial drug were identified and authenticated by the experts and were purified and the medicine was prepared as mentioned in the Siddha literature. In depth literature review was carried out in various aspect like Siddha aspect, modern aspect and scientific review were disclosed about the ingredients of the drug and it was strongly supported that possesses wound healing, anti-inflammatory and analgesic activities. The trial drug Kukkilathi Urundai was prepared in Gunapadam lab, National Institute of Siddha under the supervision of research guide and department faculties. The drug was subjected to physicochemical, chemical analysis, microbial load, HPTLC, aflatoxin and ICP-OES through standard methods. which provided the required information towards the safety and efficacy of trial drug. Acute toxicity study was carried out and the results reveled that the drug Kukkilathi Urundai have no toxic substance. The pharmacological studies, revealed that the drug Kukkilathi Urundai exhibited significant wound healing, anti-inflammatory and analgesic activities in animal models. The results of all analytical , toxicological and pharmacological studies were correlated with the available scientific information and finally concluded that the study drug Kukkilathi Urundai possess wound healing, anti-inflammatory and analgesic activities which can be used clinically for management of Kuzhippun (deep sore), Aratha viranam (non - healing ulcer), Vatha soolai (artho typhoid), Kaikaal mutakku (Rheumatic affection of limbs), Kaikaal kadupu (pain in extremites), and soolai kattu (Rheumatic gout). CONCLUSION: From the qualitative and quantitative studies, acute toxicity study and pharmacological activities (wound healing, anti-inflammatory and analgesic activities), this study is providing that the test drug Kukkilathi Urundai possess good wound healing, anti-inflammatory and analgesic activities. Finally, this research study concluded that the study drug Kukkilathi Urundai can be used for Kuzhippun (deep sore), Aratha viranam (non - healing ulcer), Vatha soolai (artho typhoid), Kaikaal mutakku (Rheumatic affection of limbs), Kaikaal kadupu (pain in extremites), and soolai kattu (Rheumatic gout).
