Abstract

BACKGROUND: Human herpesvirus 8 (HHV-8) is a DNA virus associated with the pathogenesis of Kaposi Sarcoma - a neoplasm of endothelial cells. The virus is otherwise known as Kaposi Sarcoma-associated Herpes Virus (KSHV). Kaposi Sarcoma is the most common neoplasm seen in HIV seropositive individuals. It has been reported that HHV-8 virus acquisition occurs in early childhood with an increase in infection through adulthood. AIM OF THE STUDY: To detect and quantify the expression of Human Herpes Virus 8 in a cohort of 40 HIV seropositive and 20 HIV seronegative children. MATERIALS AND METHODS: Whole mouth fluid was collected from pediatric HIV seropositive and seronegative children. DNA was extracted from the whole mouth fluid samples of HIV seropositive and seronegative children. The DNA was subjected to Real Time Polymerase Chain Reaction to detect and quantify HHV-8 in the whole mouth fluid of pediatric HIV seropositive and seronegative children. RESULTS: The presence of HHV-8 was detected and quantified using Real Time PCR technique. It was shown that HHV-8 was not detected in any of the whole mouth fluid samples of 40 HIV seropositive and 20 HIV seronegative children. CONCLUSION: HHV-8 is the most common infection in HIV seropositive children in endemic regions. In our cohort we did not detect the presence of HHV-8 in the whole mouth fluid of pediatric HIV seropositive and seronegative children which is consistent with the reported low prevalence of HHV-8 infections in India.
