Abstract

INTRODUCTION Oral squamous cel l carcinoma (OSCC) is the 11 th most common cancer in the world. Consumpt ion of tobacco and i ts products contribute for more than 90% of the OSCC. Early detect ion of OSCC at i ts precancerous stage great ly increases the chances for successful treatment . Tumor markers are proteins that are present and/or produced in high amount by mal ignant cel ls which provides informat ion on the biological behaviour of cancer- i ts aggressiveness, response to therapy and targeted treatment . Li terature confi rms that , large tumor suppressor kinase 1 and 2 (LATS1 & LATS2) are the upst ream tumor suppressor kinases in hippo signal l ing pathway that plays a significant role in oral squamous cel l carcinoma. This study was at tempted to elucidate the expression pat tern and the role of large tumor suppressor kinase 1 and 2 in the disease process of potent ial ly mal ignant disorder and oral squamous cel l carcinoma in sal iva using ELISA test . AIM To quant i fy large tumor suppressor kinase 1 and 2 in tobacco associated potent ial ly mal ignant disorder and oral squamous cel l carcinoma. MATERIALS AND METHODS This study consists of a total of 40 subjects comprising 10 normal controls (Group I) , 10 subjects of smokers wi thout lesion (Group II) , 10 subjects wi th tobacco associated potent ial ly mal ignant disorder (Group III) and 10 subjects wi th tobacco associated oral squamous cel l carcinoma (Group IV) . Sal iva samples was col lected f rom al l 40 subjects and the expression of large tumor suppressor kinase 1 and 2 were quant ified using ELISA test . STATISTICAL ANALYSIS One way ANOVA test was used to analyse the expression of LATS1 and LATS2 in study groups and also among various cytological grades. Independent sample t -test was used to analyse the expression of LATS1 and LATS2 among the histopathology grades of oral squamous cel l carcinoma subjects (Group IV) . RESULTS Group I (Normal control ) had a mean LATS1 and LATS2 value of 2.7047ng/ml and 9.9093ng/ml respect ivel y, Group II (Smokers wi thout lesion) had a mean LATS1 and LATS2 value of 3.2828 ng /ml and 7.6075ng/ml respect ivel y, Group II I (Potent ial ly mal ignant disorder ) had a mean LATS1 and LATS2 value of 4.5442ng/ml and 10.8238ng/ml respect ively and in Group IV (Oral squamous cel l carcinoma) the mean LATS1 and LATS2 value was 5.0524ng/ml and 3.0849ng/ml respect ivel y. CONCLUSION LATS1 and LATS2 proteins are sufficient ly expressed in sal iva. Its expression in sal iva can be valuable in observing the disease transformat ion from a potent ial mal ignant disorder to carcinoma. This may help in the early predict ion of oral squamous cel l carcinoma at i ts precancerous stage and also in moni toring the disease process.
